Orlistat
Orlistat is the saturated derivative of lipstatin, a potent natural inhibitor of pancreatic lipases isolated from the bacterium Streptomyces toxytricini. It is a gastrointestinal lipase inhibitor for obesity management that acts by inhibiting the absorption of dietary fats. It works by inhibiting gastric and pancreatic lipases, the enzymes that break down triglycerides in the intestine. When lipase activity is blocked, triglycerides from the diet are not hydrolyzed into absorbable free fatty acids, and instead are excreted unchanged. It is indicated for obesity management including weight loss and weight maintenance when used in conjunction with a reduced-calorie diet. It also modestly reduces blood pressure and appears to prevent the onset of type 2 diabetes. It is indicated for obese patients with an initial body mass index (BMI) ≥ 30 kg/m² or ≥ 27 kg/m² in the presence of other risk factors (e.g., hypertension, diabetes, dyslipidemia).
